He's not particularly resilient, can't fly, and doesn't have a ranged attack, so I imagine this strategy would work.
If not then Mothra can simply hover above him and release her poison powder. Sure, after Kong dies she might die as well from utilizing the attack, but Mothra will be reborn.
So either way: Mothra takes it.
