Martin Scorsese's The Stand.

I think Scorsese's proven ability to handle large casts of highly talented actors (in films like Goodfellas, Casino, The Departed) would be the best bet to bring a novel like The Stand, which has many "main" characters, to life.

A couple others:
John Carpenter's 1984
The Coen Brothers' Count of Monte Cristo
Quentin Tarantino's Frankenstein (sorry GDT fans, I just don't think he's the best man for that novel...but...)
GDT's Watership Down

and a couple projects that fell through that I always wanted to see:
Neil Blomkamp's Halo
Matt Stone and Trey Parker's kaiju film.

That'd be crazy, but I'd love to see Tarantino take on something like that....
